Abstract:In a study of 37 leprosy patients, the oral administration of levamisole failed to provoke an increase in both the Fernandez and Mitsuda reactions to lepromins of human and armadillo origin. We interpret this as evidence against an effective specific immunostimulatory capability of levamisole in leprosy patients under the conditions of the study. Current knowledge of the mechanism of levamisole action supports the concept that the fundamental immunologic defect in lepromatosus leprosy may reside in the lymphocyte and not the macrophage, or the respective related functions of these two cell forms.(AU).

Abstract:Although single-dose amphotericin B therapy appears to be immunostimulatory in mice, no data are available regarding the effects of chronic anti-fungal drug therapy on the immune system. We studied the effects on the guinea-pig cellular immune system of 4 weeks of treatment with amphotericin B, 5-fluorocytosine, or the combination of both drugs. The in vitro lymphocyte response to phytohaemagglutinin and the specific antigen, picryl human serum albumin (picHSA), were not affected by anti-fungal drug treatment. At 1.5 weeks of therapy with amphotericin B, skin test reactivity to picHSA was significantly reduced but returned toward normal by the end of 3.5 weeks of drug therapy. Macrophage migration inhibitory factor production by guinea-pig peripheral blood lymphocytes was significantly reduced after 4 weeks of amphotericin B therapy. No immunostimulatory properties could be ascribed to amphotericin B. 5-fluorocytosine had no effect on cellular immunity.(AU).

Abstract:This paper presents a review of 123 cases of leprosy of different clinical types as regards to their status of cellular immunity. These 123 cases included 41 fresh cases, 18 cases of reaction and 64 cases of leprosy taking antileprosy treatment. Out of 41 untreated cases only 11 turned up for follow up and their lymphoblastic transformation was repeated 4 to 6 months after initiating the treatment. It was observed that cell mediated immunity as expressed in terms of percentage of blast cells is definitely depressed in leprosy, most in LL and least in TT. There is a definite increase in the percentage of blast cells after taking antileprosy treatment. The rise in percentage of blast cells and hence cellular immunity is relatively more in patients treated with Lamprene as compared to those treated with DDS. Reactions also have impact over immunity in leprosy. Thus, most of the patients with ENL show higher values for blast percentage as compared to those with lepra reaction. It appears that serial lymphocyte cultures if done in all cases of leprosy undergoing treatment will help in assessment of individual progress.(AU).

Abstract:A resposta imune celular bacilo especifica na hanseniase desenvolve-se eficazmente, na grande maioria dos individuos infectados pelo Mycobacterium leprae, protegendo-os do desenvolvimento da infeccao. Numa minoria, identificam-se falhas nessa resposta imune, configurando-se verdadeiros estados de hipersensibilidade imunologica, imunodeficiencia e autoimunidade, o que provoca no hospedeiro uma serie de danos teciduais de importante morbidade clinica. O tratamento classico atualmente recomendado pela Organizacao Mundial da Saude consiste no emprego simultaneo de alguns farmacos, a denominada multidrogaterapia. Essa forma de tratamento tem resultado em melhoras satisfatorias de ordem clinica, bacteriologica e histopatologica, controlando a infeccao na maioria dos casos(AU).

Abstract:O presente trabalho teve como objetivo a avaliaçao do estado imunitário celular em pacientes com hanseníase virchoviana, em nosso meio, através de reaçoes cutâneas de leitura tardia. Além disso, procuramos definir e delimitar o valor dessas reaçoes individualmente e em conjunto. Concomitantemente ao estudo da imunidade celular nos pacientes "virchovianos", foi realizado o mesmo estudo em em grupo de indivíduos "normais". Nessa avaliaçao utilizamos uma bateria de reaçoes cutâneas constituída de antígenos específicos (levedurina, tricofitina, PPD e Escherechia coli), de uma substância capaz de induzir sensibilizaçao de contato (DNCB)* e da PHA, por via intradérmica, a qual independe de sensibilizaçao prévia. Foram estudados 95 pacientes "virchovianos" e 30 indivíduos "normais". As reaçoes cutâneas e a leitura das mesmas foram sempre realizadas pelo mesmo indivíduo. Foram utilizadas as mesmas seringas para um mesmo antígeno. Cada paciente foi submetido a uma reaçao intradérmica de cada vez(AU).
