Abstract:Visando contribuir para o melhor entendimento da participação das citocinas na hanseníase dimorfa, o presente estudo investigou a produção desses mediadores in vitro e in situ em pacientes dimorfos-tuberculóides (HDT) e dimorfos-virchovianos(HDV). Foram avaliados 7 pacientes HDT e 12 HDV, virgens de tratamento, além de 19 indivíduos sádios (grupo controle). Culturas de células mononucleares do sangue periférico (PBMC) foram estimuladas ou não com estímulos inespecíficos e específicos do M. Leprae (antígeno inteiro e sonicado) e após 48 horas o sobrenadante foi recolhido para dosagens das citocinas TNF-a, IFN-y, IL-10 e TGF-B1. Biópsias das lesões cutâneas foram submetidas aos procedimentos histológicos por meio da coloração com hematoxilina-Eosina e Faraco-Fite; os cortes foram submetidos, ainda, à detecção in situ de iNOS, IL-10 e TGF-B1 por imunoistoquímica. A quantificação de citocinas em sobrenadante de PBMC revelou que pacientes HDT produziram níveis menores de TGF-B1 e pacientes HDV, níveis menores de IL-10...(AU)^ipt.

Title:Behavior of leprosy bacilli in complex liquid media with highly available sources of nutrient and acessory substances.

Abstract:Completely homogenized suspensions of fifteen leprous nodules were inoculated into complex natural media such as blood, serum, leprous tissue extracts, chick embryo juice, and sterile egg yolk suspensions. These substances were supplemented with a variety of proteolytic digests, autolysates, peptones, products derived from acid-fast bacilli, yeast extract and simpler sources of nitrogen and carbon. Each inoculated medium also contained all of the nutrient substances present in the lepromata. Diphtheroid bacilli were recovered from three of six cases showing ulcers of the skin. One culture of a streptothrix and two of acid-fast bacilli were also obtained from one of the ulcerated cases. A third acid-fast microorganism was cultivated from a patient who showed no ulcers. These acid-fast cultures were not tubercle bacilli. By quantitative microscopic studies in liquid media from which aliquot portions were removed for comparison with control smears or formalinized suspensions, it was learned that the leprosy bacilli did not multiply in any of the 109 nutritional combinations tested, or in the presence of added carbon dioxide and oxygen. Growth did not occur in media shown to be suitable for the cultivation of human blood monocytes or of fibroblasts from lepromata. (AU).

Abstract:In some subjects the infective agent of leprosy, Mycobacterium leprae, causes disseminated (lepromatous) disease. Such subjects have a major role in the transmission of the disease and show deficient T-cell responses both in vivo and in vitro to M. leprae, but not to other antigens. Numerous studies have recently shown that T cells with functional capabilities after initial triggering with antigen can be maintained in a state of continuous proliferation in vitro when cultured in medium containing interleukin 2 (IL-2). Here we have studied the effect of IL-2 rich T-cell conditioned medium on lepromatous peripheral blood mononuclear cells. Our results show that although lepromatous T cells fail to produce IL-2 after exposure to M. leprae they can respond by proliferation to M. leprae in the presence of T-cell conditioned medium, suggesting that the unresponsiveness in lepromatous leprosy results from a deficiency in the production of IL-2 or related factors and not a lack of M. leprae-reactive T cells.(AU).

Abstract:Mixed leucocyte cultures, from two normal donors, were set up in media containing human serum from one of the following sources: (a) a pool of normal group AB donors; (b) Chinese, Malay or Indian patients with untreated leprosy; (c) the same patients after effective anti-leprosy treatment; (d) control Chinese, Malay or Indian subjects. Transformation was estimated by measuring the incorporation of tritiated thymidine in the last 24 hr of a 7-day culture period. Transformation was impaired in sera from treated lepromatous patients, but was less impaired or not impaired at all in sera from treated lepromatous patients. The loss of depressive activity after treatment was more marked in Chinese and Indian than in Malay patients. Transformation was also impaired, though to a lesser extent, in sera from patients with untreated tuberculoid leprosy; it was still impaired in sera from treated tuberculoid patients. There was no evidence of specificity in impairment of mixed lymphocyte reactivity and lymphocytotoxic antibodies appeared to play no role. The incidences of hepatitis B antigen and antibody and of anti-nuclear factor were not notably high.(AU).

Abstract:Cultivation trials for Mycobacterium leprae resulted in growth of Mycobacterium psychrophilum (L). Media were inoculated with host grown Mycobacterium leprae cells from armadillo tissues, Nu mice foot pads or human lepromata. Cultures were obtained in liquid and on semisolid multifactoria 1 media containing water soluble palmitic acid or its salts. Ammonium thioglycolate and Napalmitate served as carbon and energy sources. The water soluble palmitic acid remained in perfect solution following sterilization in the autoclave, thus easily accessible to the cells. The cyclodextrin-Fe complex served as a siderophore to grow the obtained leprosy derived psychrophilic cells. The leprosy derived cultures and subcultures grew opimally at+10 degrees Celssius but deteriorated rapidly at + 32 degrees Celsius, in the multifactorial media. No growth occurred in 7H9 media...(AU).
