Abstract: Resumo: In this work we describe the purification and characterization of armadillo immunoglobulins. The IgM was precipitated using low-strength ionic solution and further purified by filtration through Sephadex G-200. The IgG was obtained in pure form by precipitation of serum with ammonium sulfate and DEAE-cellulose ion exchange chromatography. The purity of these immunoglobulins was evaluated by polyacrylamide gel electrophoresis. The results showed 28-kDa light chains and 55-kDa and 70-kDa heavy chains for IgG and IgM, respectively. The rabbit antibodies against these molecules were used to prepare fluorescein (FITC) and peroxidase conjugates. The FITC conjugate was used to quantify IgM-bearing lymphocytes. An average of 17% of peripheral blood lymphocytes were sIgM+ from 14 healthy animals. Additionally, in the same animals we quantified lymphocytes with the capacity to form rosettes with sheep red-blood cells; the average for this marker was 10%. Also, the production of crossreacting antibodies to BCG was evaluated in healthy and Mycobacterium leprae-inoculated animals using the peroxidase conjugates. All animals with active infection recognized BCG antigens.

Abstract:Secondary amyloidosis is the leading cause of death in lepromatous leprosy in this country. The mortality is directly caused by amyloid involvement of the kidneys, which occurred in 100% of this series. The diagnosis of this complication depends largely on the development of a persistent proteinuria, Congo red absorption, hepatosplenomegaly and, to a lesser degree, altered serum proteins and perhaps liver biopsy in selected cases. the correlation between the Congo red test and the degree of amyloidosis is poor, and unless this test shows 80 to 100% dye absorption in one hour the diagnosis cannot be made from this finding alone. Serum protein and lipoid changes occur in lepromatous leprosy per se, and in a study of the various protein fractions no consistent alteration occurred in cases complicated by amyloidosis. The average duration of life after the onset of proteinuria in amyloidosis was 38.33 months, with a fairly wide range above and below this figure. The development of anemia and a progressive elevation of the nonprotein nitrogen are unfavorable prognostic signs.

Abstract:Leprosy presents as a clinical and immunological spectrum of disease. With the use of gene expression profiling, we observed that a distinction in gene expression correlates with and accurately classifies the clinical form of the disease. Genes belonging to the leukocyte immunoglobulin-like receptor (LIR) family were significantly up-regulated in lesions of lepromatous patients suffering from the disseminated form of the infection. In functional studies, LIR-7 suppressed innate host defense mechanisms by shifting monocyte production from interleukin-12 toward interleukin-10 and by blocking antimicrobial activity triggered by Toll-like receptors. Gene expression profiles may be useful in defining clinical forms of disease and providing insights into the regulation of immune responses to pathogens. (AU).

Abstract:Los resultados de mayor interés obtenidos sobre las muestras de LCD y suero de 7 casos de hanseniasis (H.D.) (2 borderline: B 3 lepromatosos: L de los cuales el 4 L grave y con déficit inmunitario; 2 tuberculoides: T), son: I.º) Los índices LCR/suero de la albúmina y de las IgG por tests cinético-nefelometría y de las IgG anti PGL-1 por ELISA, en cuanto separan los casos según sus tipos y forma mediante valores típicamente uniformes: los más bajos de albúmina para los L; los más bajos de IgG anti PGL-1 para los B; los medios de IgG para los T, siendo los B y L muy deformantes. II.º) Los index de LINK indican producción intratecal de IgG si superan el V.N. en nuestros casos los de IgG anti PGL-1 han resultado todos superiores y altos, sólo los B menos altos; los IgG todos inferiores; pero, hay que considerar que por la policlonía de la IgG, una parte de ellas podría resultar superior si fuese revelada aisladamente por su antígeno específico. III.º) Una mayor casuística con test de medidas homólogas para los dos de IgG podrá confirmar o no nuestros resultados, y también modificar nuestra de ducción por la cual los epítopes HLA-DR de los queratinocitos uniéndose con las IgG intratecales determinarían la respuesta al test lepromínico, es decir, negativa en los L por la falta de los HLA-DR; positiva la precoz en los B po la unión con la IgG; positiva la tardía y a menudo la precoz en los T por la unión con la IgG anti PGL-1 y las IgG. Otro caso de diagnosis incierta resulta positivo para la H.D. de tipo T, pero atípico o la debido a la asociación de otra enfermedad del S.N.C. como la esclerosis múltiple o la siringomielía. (AU).

Abstract:Sequential estimates of the levels of circulating immune complexes (CIC), complement catabolic fragment C3d, complement-mediated immune complex solubilization (CMS) and immunoglobulins were made in 24 newly diagnosed with borderline tuberculoid leprosy over a 20 month period after initiation of chemotherapy. Fourteen of these patients had not suffered from reversal reactions either at the time of presentation or during the follow-up. The levels of CIC were evaluated in them from the third to the eleventh month after starting chemotherapy and immunoglobulin G (IgG) levels were evaluated up to eight months. The concentrations of C3d and immunoglobulins A (IgA) and M (IgM) were normal in these patients. The other ten patients had reversal reaction at the time of diagnosis which subsided by the third month after starting treatment. They did not have reversal reactions later. The levels of CIC and IgG were elevated and those of CMS were depressed throughout the study period. Serum C3d level was initially elevated but came down to normal by the third month while IgA and IgM levels were within normal limits. The relevance of these findings to the genesis of reversal reaction is discussed in this communication.(AU).

Abstract:About twenty distinct antigenic components have been demonstrated in Mycobacterium leprae (M. leprae) by crossed immunoelectrophoresis against a rabbit antiserum produced by immunization with concentrated M. leprae antigen. This system allows a more detailed analysis of the antigenic relationship between M. leprae and other mycobacteria and a better characterization of the antigenic content of various M leprae preparations than with previously available anstisera of the antigenic content of various M. leprae preparations than with previously available antisera which reacted with far fewer components. The antibody activity in sera of patients with lepromatous leprosy was studied by incorporating the sera into the intermediate gel of the M. leprae reference system. Antibodies were found against only seven of the components. Since those compared are all known to be cross-reacting widely with antigens of other mycobacteria, it is speculated that cross-immunization may influence the antibody response in lepromatous leprosy.(AU).
