Abstract:A CENTURY-LONG search for an unaltered animal in which to study leprosy in humans culminated in the finding that the nine-banded armadillo (Dasypus novemcinctus, Linn.) a primitive mammal native to the southern Western Hemisphere, develops disseminated lepromstous leprosy following inocuation with Mycobacterium leprae isolated from human tissue.(AU).

Abstract:Although single-dose amphotericin B therapy appears to be immunostimulatory in mice, no data are available regarding the effects of chronic anti-fungal drug therapy on the immune system. We studied the effects on the guinea-pig cellular immune system of 4 weeks of treatment with amphotericin B, 5-fluorocytosine, or the combination of both drugs. The in vitro lymphocyte response to phytohaemagglutinin and the specific antigen, picryl human serum albumin (picHSA), were not affected by anti-fungal drug treatment. At 1.5 weeks of therapy with amphotericin B, skin test reactivity to picHSA was significantly reduced but returned toward normal by the end of 3.5 weeks of drug therapy. Macrophage migration inhibitory factor production by guinea-pig peripheral blood lymphocytes was significantly reduced after 4 weeks of amphotericin B therapy. No immunostimulatory properties could be ascribed to amphotericin B. 5-fluorocytosine had no effect on cellular immunity.(AU).

Abstract:The response to soluble mycobacterial antigens of peripheral blood mononuclear cells, from most normal donors, tuberculosis patients or cases of tuberculoid leprosy (TT/BT) was enhanced by the addition of indomethacin. In contrast, indomethacin caused no enhancement of the response of cells from lepromatous leprosy (BL/LL) cases. Moreover the addition of 10-5M prostaglandin E2 (PGE2) failed to inhibit the proliferative responses of cells from the BL/LL patients, although it markedly inhibited the responses of peripheral blood mononuclear cells from the other groups. The addition of PGE2 or indomethacin to cells which had been precultured for 48 hours had no significant effect on the proliferative responses of cells from any of the groups of donors. These results suggest that a normal, prostaglandin-dependent, indomethacin-sensitive regulatory mechanism is absent from the peripheral blood mononuclear cells of BL/LL patients.(AU).

Abstract:The effects of dapsone on polymorphonuclear leukocyte functions and lymphocyte mitogen-induced transformation were assessed in vitro and in vivo in normal individuals and in newly diagnosed untreated patients with lepromatous leprosy. The effects of dapsone on the cell-free generation of superoxide by the xanthine: xanthine oxidase system and iodination of bovine serum albumin by horseradish peroxidase were also investigated. In normal individuals dapsone mediated stimulation of polymorphonuclear leukocyte migration in vitro and vivo. Dapsone had no effect on postphagocytic hexose monophosphate shunt activity in vivo. Similar effects were found in patients with lepromatous leprosy. Dapsone also decreased the inhibitory activity of serum from patients with lepromatous leprosy on normal polymorphonuclear leukocyte migration in vitro. Progressive loss of serum-mediated inhibition of migration was observed after ingestion of dapsone by the patients. Further experiments showed that stimulation of polymorphonuclear leukocyte motility was related to inhibition of lymphocyte transformation at high concentrations in vitro, but had slight stimulatory activity on phytohemagglutinin-induced transformation in controls and patients in vivo.(AU).
