Abstract:Four soluble antigens of Mycobacterium leprae have been identified using 12 murine monoclonal antibodies. Their specificity, taxonomic distribution and molecular nature were analysed by radioimmunoassays and by immunoblotting from polyacrylamide electrophoresis gels. A protein antigen MY1 (12K) reacted with one antibody (ML06) without demonstrable cross-reactivity for any of the other 20 tested species of mycobacteria. Another four antibodies which identified antigen MY2 revealed only a marginal degree of cross-reactivity with three other species of mycobacteria. Two antigens shared by several other mycobacteria species were: MY3 represented by five protein bands (35-70K) and a subtilisin resistant molecule MY4 (40-50K) with two distinct determinants and presumably of polysaccharide nature. The described monoclonal antibodies may represent novel valuable diagnostic reagents as well as tools for the purification of antigens which could be explored towards prophylactic or therapeutic immunization against leprosy.(AU).

Abstract:All mycobacteria species share some antigens, so there may be cultivable mycobacterial cultures that can provide vaccine protection against leprosy. Vaccine protection against Mycobacterium leprae infections in mice has been demonstrated for M. leprae itself, as living or heat-killed suspensions, and for Mycobacterium bovis (BCG), as living suspensions. Results are reported here with 17 other cultures. The mycobacterial suspensions were injected intradermally, and the mice were challenged in the footpad with infectious suspensions of M. leprae. In two experiments the mice were also challenged by footpad injections of 10(7) heat-killed M. leprae so the footpad enlargment could be measured. That some mycobacterial suspensions were immunogenic for some of their own antigens was suggested by reactions at the vaccine site and enlargement of the regional lymph nodes. Some mycobacterial suspensions also stimulated footpad enlargement on challenge by homologous suspensions or by challenge with M. leprae suspensions. Consistent protection against infectious challenge with M. leprae was observed only with BCG and M. leprae, however...(AU).

Abstract:The response to soluble mycobacterial antigens of peripheral blood mononuclear cells, from most normal donors, tuberculosis patients or cases of tuberculoid leprosy (TT/BT) was enhanced by the addition of indomethacin. In contrast, indomethacin caused no enhancement of the response of cells from lepromatous leprosy (BL/LL) cases. Moreover the addition of 10-5M prostaglandin E2 (PGE2) failed to inhibit the proliferative responses of cells from the BL/LL patients, although it markedly inhibited the responses of peripheral blood mononuclear cells from the other groups. The addition of PGE2 or indomethacin to cells which had been precultured for 48 hours had no significant effect on the proliferative responses of cells from any of the groups of donors. These results suggest that a normal, prostaglandin-dependent, indomethacin-sensitive regulatory mechanism is absent from the peripheral blood mononuclear cells of BL/LL patients.(AU).

Abstract:Guinea-pigs were immunized with Mycobacterium leprae in saline and with autoclaved preparations of Mycobacterium w., ICRC bacillus, Mycobacterium phlei and Mycobacterium vaccae. A group of animals were also immunized with live Mycobacterium w. All animals were challenged after one month of M. leprae and other mycobacteria. Induration produced in response in guine-pigs to challenge with homologous mycobacterial preparations and M. leprae. In most cases, the early reaction was higher with homologous antigens as compared to M. leprae antigens were however of comparable order especially in animals immunized with Mycobacterium w, and ICRC bacillus. Animals immunized with M. leprae gave low late reactions with preparations from Mycobacterium phlei and Mycobacterium vaccae.(AU).

Abstract:Skin reactions have been measured in tuberculoid and lepromatous leprosy patients with Dharmendra and Mitsuda type of lepromins prepared from M. leprae, Mycobacterium w, ICRC bacillus, M. phlei, M. vaccae and M.gordonae. In tuberculoid patients Mycobacterium w gave the closest response to M. leprae, however, in lepromatous and borderline lepromatous cases, this bacteria produced greater response than M. leprae. (AU).

Abstract:18 cases of different types of leprosy were studied to see the skin reactions to known lepromin and to antigens from four cultivable saprophytic mycobacteria. ICRC bacillus gave best results in tuberculoid leprosy - even as campared to M. leprae, whereas M. delhi (M.W.) with M. leprae is concerned, there was hardly any difference between the two.(AU).
