Resumo:Immunoblot assays for the antibodies to Mycobacterium tuberculosis sonic extracts showed that all serum specimens of 40 lepromatous and 28 tuberculoid leprosy patient reacted in a significant amnner to 29/33-kilodalton (kDa) doublet and 64-kDa antigens, respectively. By using an enzyme-linked immunosorbent assay, we observed a significantly high immunoglobulin G antibody titer to the purified M. tuberculosis 29/33-kDa doublet and 64-kDa antigens in lepromatous and tuberculoid leprosy pateints, respectively, as compared with normal subjects and tuberculosis patients. This enzyme-linked immunosorbent assay serology may be useful for distinguihing two polar types of leprosy and for diagnosing leprosy in general (AU).

Resumo:Humans are exposed worldwide to a variety of environmental mycobacteria (EM) and most children are inoculated with live Bacille Calmette-Guérin (BCG) vaccine. Although rarely pathogenic, poorly virulent mycobacteria, including BCG and most EM, may cause a variety of clinical diseases. M. tuberculosis and M. leprae are more virulent, causing tuberculosis, and leprosy, respectively. Remarkably, only a minority of individuals develop clinical disease, even if infected with virulent mycobacteria. There is now accumulating evidence that the large interindividual variability of clinical outcome results in part from variability in the human genes that control host defense. We review here in current knowledge about genetic predisposition to common (leprosy and tuberculosis) and rare (BCG and EM infections) mycobacterial infections. (AU).

Resumo:The commonest pathogenic skin mycobacteria in many areas of the world is still Mycobacterium leprae. This mycobacteria, which grows best at skin temperatures 31(graus)C-33(graus)C, often involves nerve twigs and trunks. In addition to producing skin lesionms, it can cause motor and sensory deficits. The mycobacteria has been cultivated in the laboratory only in animal models.(AU).

Resumo:At varying periods of time following the successful establishment of systemic infections with Mycobacterium leprae or M. lepraemurium in the mouse and the nine-banded armadillo eyes were examined by light microscopy. Inoculation of bacilli was by the intravenous or intraperitoneal route or directly into the hind footpads; eyes were not directly inoculated in this study. During periods of up to 3 years under laboratory conditions no animal showed evidence of impaired vision or blindness, and the external appearance of both eyes was normal. The ocular histopathology and the sites of accumulation of bacilli are described. In immunologically normal mice infected with M. lepraemurium bacilli were much commoner in extraorbital tissues, but they were, nevertheless, found in various tissues within the orbit, including the ciliary body and sclera. In immunologically normal mice (and one rat) injected with M. leprae of human origin no bacilli were found in the eye, but in mice immunologically depressed by thymectomy and total body irradiation considerable numbers of bacilli were present in the iris and ciliary body and also in the limbal cornea. In the armadillo bacilli were found in large numbers in virtually all tissues except the lens, retina, optic nerve, and aqueous and vitreous humours, but the uveal tract was heavily involved. Findings are discussed in relation to the great frequency of ocular involvement and the importance of immune-complex disease in patients with lepromatous leprosy, and to factors wihch may favour the localisation and multiplication of Mycobacterium leprae in the eye.(AU).

Resumo:Two men with progressive synovitis and osteomyelitis of the wrist were found to have infection with Mycobacterium fortuitum. Immunological defects were demonstrated in both cases in association with renal failure and renal transplantation respectively. Failure of appropriate antibiotic therapy to eradicate infection necessitated amputation in one case. M. fortuitum is frequently sensitive to only amikacin or kanamycin. Treatment with amikacin alone was unsuccessful. This organism is widespread in the environment and may need to be considered in undiagnosed chronic infection in the immunologically suspectible host, as it poses unusual problems in diagnosis and therapy.(AU).

Resumo:Neonatal thymectomy and neonatal thymectomy plus ATS resulted in a merked increase in susceptibility of both Buffalo and Lewis rats to infection with M. leprae. In Buffalo rats, increase in susceptibility was limited to footpad infection. In Lewis rats, this was extended to include testis infection where the organisms in thymectomized-ATS-treated rats were found to be still in the logarithmic phase of growth 15 months after inoculation.(AU).

Resumo:18 cases of different types of leprosy were studied to see the skin reactions to known lepromin and to antigens from four cultivable saprophytic mycobacteria. ICRC bacillus gave best results in tuberculoid leprosy - even as campared to M. leprae, whereas M. delhi (M.W.) with M. leprae is concerned, there was hardly any difference between the two.(AU).
